It was a nice article to understand the points to be considered in validating an autoclave. But there are some questions in my mide, please clarify that.

  1. Firstly it is ok to reduced the number of probes when you ahve atight schedule like the one in production autoclave.
  2. It is viable to revalidate the probes if you are conutinouly using them in the autoclave validation run. (this is will be used in QC autoclave.)

Thanks

Sameer Pandit.

Nice article,

Thank you

nice article, keep posting such kind of articles on validation studies on fermentors and inactivation tanks

The article is very nice,
I would like to know the biological indicator which is used for the validation of autoclve when it is run at 115 degree centigrdae temperatute for 15 minute.
Thanks

Dear R.C. Pantola

Before using BI first of all you have to check the D value on 115 C temperature if suppose D value on 115 2.5 min. then you desided how many log reduction you want if you want to go for 8 log reduction then

8 x 2.5 = 20 min

You have to run your autoclave for 20 minute at 115 C that is why you can not keep the time verbely.

B. stearothermophillus have the D value on 121 C not 115 only B.subtillis 5383 have the D value on 106 or 110 C Temperature this is called the low temperature sterilization.

Dear Chudhary
My query was not for d value, f valu and for how long it has to be run.It was for which one has(BI) has to be selected.
Thanks
