I am having Acetaminophen Tablets 100 mg, for that i am doing assay method
As a rule of thumb i have take both standard and samples solution at the same concentration.
As per USP i have make 0.01mg/ml solutions of both Standard and sample.
standard solution[0.01mg/ml] : 10 mg of standard to 100 ml mobile phase [0.1mg/ml]. Then 1 ml of 0.1mg/ml solution to 10ml with mobile phase[0.01 mg/ml].
Then my question is how to prepare 0.01mg/ml sample solution.
Pleas help me

Hi friend,
For this you should know the single tablet weight( API+ Excipients)
Take 20 tablets and grind to powder.
Now take an amount that contains 10 mg of API to 100 ml then 1 ml to 10 ml dilute with mobile phase.
Eg: you have a tablet claiming 100 mg of Acetaminophen [ single tablet contins 100 mg of API] and the tablet weight is 200 mg(you have not written the each tablet weight, I assumed 200 mg for each tablet weight].
Sample preparation : 20 mg of grinded powder(It contains 10 mg of API) dissolved in 100 ml, further diluted1 ml to 10 ml.
