Microbiological method validation for probiotic (dietary suplement)

Hello Dear members of forum,

Do anyone have suggestions regarding the microbiological method validation for probiotics. My product contains four bacterial strain (L.acidophilus, L. rhamnosus,S.thermophilus, L.bulgaricus) for analyses I use two dilutions 6 and 8, according specification product contains 2*109 probiotic bacterial count. I’m confused, I don’t know what where to start, should I use standard bacterial strains of lactobacteria for investigation and use such approach as we have during the microbiological method validation. If someone have experience in this filed or some advice, I’ll be glad to share your opinions.

Use ATCC cultures for identification. Lactobacillus is anaerobe so you will have to use a ‘Carbon Dioxide gas pack (BBL?)’. Media must be specific for your microbes (L-Media?). Follow and adapt USP <1224>, <1225>, <1226>.

Demonstrate your method is appropriate by spiking and recovering from your dietary supplement. Check NF for maximum number of CFU.
