Method validation

Dear all

In a validated method can i change the diluent ratio.

My procedure is as follows in sample add 25 ml diluent with shanking & than make the volume with water. i have done the validation with this method.

Now during analysis my peak is splitted & the reason for that is diluent.
I want to use 2.5 ml diluent instead of 25 ml.

Can i change this.

splitted peak is not due to diluent effect. you have got problem with your column.
