Media Fill Validation

Dear Sir,

My current process is having 24 hrs. hold time after filtration and before filling.

Now, we are conducting the media fill validation, so is it really necessary to hold the media for 24 hrs.

And what shall I take the intervention during the media fill.

Is it necessary to incubate the intervented units during media fill. And shall I count this units in acceptance criteria i.e. result.
